โรคติดเชื้อในปลานิล สเตรปโตคอคโคซิส 2 เชื้อแบคทีเรียสเตรปโตคอคคัส (Streptococcus spp.) ที่ก่อปัญหาในปลาในปลาจัดเป็นแบคทีเรียในวงศ์ Streptococcaceae เมื่อย้อมสีแกรมแล้วนำมาส่องดูลักษณธของเชื้อโดยใช้กล้องจุลทรรศน์จะพบลักษณะเชื้อติดสีแบคทีเรียแกรมบวก ขนาดเล็ก เส้นผ่านศูนย์กลาง 0.5-1 ไมโครเมตร รูปร่างกลมหรือรูปไข่ มีการเรียงตัวของเซลล์เป็นสายโซ่ (cocci chain) ซึ่งมีความยาวแตกต่างกันตั้งแต่ต่อกันคู่เดียว จนถึงมากกว่า 30 เซลล์ ทั้งนี้ขึ้นกับชนิดของเชื้อและสภาพแวดล้อมเชื้อแบคทีเรียสเตรปโตคอคคัสเจริญเติบโตได้ดีในอาหารเลี้ยงเชื้อแบคทีเรียที่มีสารอาหาร (enriched bacteriologic media) เช่น เนื้อเยื่อจากสัตว์ ซีรัม หรือเลือดที่ผสมในอาหารเลี้ยงเชื้อ เชื้อนี้มีคุณสมบัติทำให้เกิดการแตกตัวของเม็ดเลือดแดง (hemolysis) 2 ลักษณะ คือ การแตกตัวแบบสมบรูณ์ (beta-hemolysis) และไม่สมบรูณ์ (alpha-hemolysis) ซึ่งอาหารเลี้ยงเชื้อที่ใช้ทั่วไปได้แก่ อาหารเลี้ยงเชื้อผสมเลือด (blood agar) เลือดสัตว์ที่นิยมใช้ในการเลี้ยง คือ เลือดแกะ เนื่องจากเห็นการแตกของเม็ดเลือดชัดเจน (streptococcal hemolytic patterns) ลักษณะของเชื้อแบคทีเรียสเตรปโตคอคคัสที่พบเมื่อเพาะบนอาหารเลี้ยงเชื้อ จะพบลักษณะเชื้อขนาดเล็กเส้นผ่านศูนย์กลางประมาณ 1-2 มิลลิเมตร พื้นผิวเรียบ โค้งมน สีขาวทึบ และมักพบการแตกตัวของเม็ดเลือดแดงรอบๆ เชื้อ คุณสมบัติทางชีวเคมีที่สำคัญของเชื้อมีหลายคุณสมบัติ เช่นไม่สามารถสร้างสปอร์ (non-spore forming) ไม่เคลื่อนที่ (non-motile) สามารถเจริญได้ทั้งภาวะมีและไม่มีออกซิเจน (หลายชนิดเจริญเติบโตได้ดีเมื่อมีคาร์บอนไดออกไซด์แค่ 2-10%) ไม่สร้างเอนไซม์คะตะเลส (catalase) ผลิตเอนไซม์เพอรอกซิเดส (peroxidase) สามารถย่อยน้ำตาลได้หลายชนิดและผลิตกรดแลกติก (lactic acid) เชื้อแบคทีเรียสเตรปโตคอคคัสไม่ทนต่อความแห้ง ความร้อนและยาฆ่าเชื้อ เช่น 70% เอทานอล ฟอร์มาลดีไฮด์ กลูตาราลดีไฮด์ และไอโอดีน เนื้อหาวันนี้ค่อนข้างลึกและเป็นวิชาการมากไปนิดหนึ่งหวังว่าจะเป็นประโยชน์สำหรับผู้สนใจนะคะซึ่งคุณสมบัติที่กล่าวมาเหล่านี้ล้วนมีผลต่อวิธีการเกิดโรคความรุนแรงของโรค ลักษณะอาการที่พบกรณีที่มีการติดเชื้อซึ่งจะกล่าวต่อไปในครั้งหน้าค่ะ สัตวแพทย์หญิง ดร.มินตรา ลักขณา (หมอเม)
Infectious disease in tilapia: Streptococcosis 2 Streptococcus spp. causing disease in fish are bacteria in family Streptococcaceae. They are small Gram-positive bacteria, with the size of 0.5-1 micrometer. They are round to oval shape, and arrange themselves into chain as known as “cocci chain”. The length of cocci chain can be from 2 cells up to 30 cells, depending on bacteria species and environment. Streptococcus can grow well in enriched bacteriologic media that made of animal tissue, serum or blood. One of the characteristics of these bacteria is the ability to lyse red blood cell. Sheep blood is often used to culture Streptococcus because it clearly presents streptococcal hemolytic patterns. Streptococcus colonies on blood agar are 1-2 mm in diameter, whitish-smooth surface. Lysis of red blood cell around the colonies are usually observed (hemolysis). For other important biochemical characteristics, Streptococcus are non-spore forming, non-motile, facultative anaerobe bacteria (some species can grow well when 2-10% CO 2 is presented). They can utilize many types of sugar. They can produce peroxidase enzyme, and lactic acid but not catalase enzyme. This is an important part! Streptococcus are not resistant and can be killed by dryness, heat and many disinfectants such as 70% ethanol, formaldehyde, glutaraldehyde and iodine. Today topic might be a little complicated.
