โปรไบโอติกกับเชื้อสเตรปโตคอคคัส มีหลายงานวิจัยที่ค้นคว้าและศึกษาเกี่ยวกับประสิทธิภาพของโปรไบโอติกต่อการต้านเชื้อก่อโรคในสัตว์น้ำ ซึ่งรวมถึงเชื้อสเตรปโตคอคคัสที่ก่อโรคในปลานิลด้วย ยกตัวอย่างเช่นงานวิจัยของนักวิจัยจากประเทศอินโดนีเซียที่ทดลองให้โปรไบโอติกหลายๆ ชนิดผสมอาหารในปริมาณที่เหมาะสม ในงานวิจัยชิ้นนี้เลือกใช้โปรไบโอติก 3 ชนิด คือ Bacillus cereus เชื้อ Bacillus subtillis และเชื้อ Staphylococcus lentus ผสมกันหลายรูปแบบในความเข้มข้นต่างๆ กัน ให้ปลานิลน้ำหนักประมาณ 13 กรัม/ตัว กินพร้อมอาหาร เป็นเวลาติดต่อกัน 14 วัน โดยแบ่งเป็นกลุ่มการทดลอง 6 กลุ่ม ดังนี้ 1. กลุ่มที่ให้อาหารผสมเชื้อ Bacillus cereus และเชื้อ Bacillus subtillis ปริมาณ 1,000,000 CFU/mL 2. กลุ่มที่ให้อาหารผสมเชื้อ Bacillus subtillis และเชื้อ Staphylococcus lentus ปริมาณ 100,000 CFU/mL 3. กลุ่มที่ให้อาหารผสมเชื้อ Bacillus cereus และเชื้อ Staphylococcus lentus ปริมาณ 100,000 CFU/mL 4. กลุ่มที่ให้อาหารผสมเชื้อ Bacillus cereus และเชื้อ Bacillus subtillis และเชื้อ Staphylococcus lentus ปริมาณ 1,000,000 CFU/mL 5. กลุ่มที่ให้อาหารธรรมดาไม่ได้ผสมเชื้อโปรไบโอติก (กลุ่มควบคุมผลบวก) 6. กลุ่มที่ให้อาหารธรรมดาไม่ได้ผสมเชื้อโปรไบโอติก (กลุ่มควบคุมผลลบ) ในวันที่ 15 ของการทดลอง นักวิจัยทดลองนำเชื้อสเตรปโตคอคคัส (Streptococcus agalactiae) ที่ก่อโรคไปฉีดเข้าในตัวปลานิลในปลากลุ่มทดลองกลุ่มที่ 1-5 (กลุ่มที่ 6 ไม่ได้ทดลองฉีดเชื้อเพราะใช้เป็นกลุ่มควบคุม) ผลการศึกษาพบว่า ปลากลุ่มที่ให้กินอาหารผสมเชื้อโปรไบโอติกทุกกลุ่มมีการตอบสนองต่อเชื้อสเตรปโตคอคคัสที่ฉีดให้ โดยมีอัตราการรอดชีวิตระหว่าง 85-89% ใกล้เคียงกับปลากลุ่มที่ 6 (88%) ในขณะที่ปลาทดลองกลุ่มที่ 5 มีอัตราการรอดชีวิตเพียง 66% เท่านั้น นอกจากนี้เมื่อตรวจการตอบสนองของระบบภูมิคุ้มกันโดยวัดจากค่า respiratory burst activity ยังพบการเพิ่มการทํางานของเซลล์ที่ทำลายเชื้อโรคเพิ่มมากขึ้น เช่นเดียวกันเมื่อตรวจดัชนีการจับกินสิ่งแปลกปลอมก็พบว่ามีค่าสูง นอกจากนี้ ยังพบการศึกษาในบราซิล เมื่อปี 2560 ที่ทดลองผสมเชื้อโปรไบโอติก Lactobacillus plantarum (เป็นโปรไบโอติกกลุ่มแลคติกแบคทีเรีย) ให้เป็นอาหารเสริมกับปลานิลและประเมินผลที่พบต่อสมรรถภาพการเจริญเติบโตการตอบสนองทางภูมิคุ้มกันรวมทั้งลองตรวจลำไส้เล็กของปลานิลที่ลองทำให้ป่วยด้วยเชื้อ Streptococcus agalactiae ด้วยกล้องจุลทรรศน์อิเล็กตรอนแบบส่องผ่าน (Transmission Electron Microscopy, TEM) โดยในการทดลองนักวิจัยได้แบ่งปลาเป็น 2 กลุ่ม คือ กลุ่มควบคุมและกลุ่มที่ได้รับอาหารเสริมด้วยเชื้อโปรไบโอติก Lactobacillus plantarum เป็นระยะเวลาติดต่อกันนาน 58 วัน ทีมวิจัยสังเกตถึงปริมาณเชื้อโปรไบโอติก Lactobacillus plantarum ที่เพิ่มจำนวนขึ้น ขณะเดียวกันกลับพบว่าปริมาณเชื้อกลุ่ม Vibrionaceae ในปลาที่ได้รับอาหารเสริมลดลงรวมถึงพบว่าปลากลุ่มทดลองให้อาหารผสมโปรไบโอติกมีน้ำหนักที่เพิ่มขึ้นอย่างมีนัยสำคัญ อัตราการเจริญเติบโตและประสิทธิภาพการใช้อาหารดีนอกจากนี้หลังจากที่ลองทำให้ปลานิลป่วยด้วยเชื้อ Streptococcus agalactiae แล้วพบว่าปลากลุ่มที่ได้กินอาหารผสมโปรไบโอติกมีปริมาณเม็ดเลือดชนิด thrombocytes และ neutrophils เพิ่มมากขึ้นด้วยซึ่งมีผลต่อการต่อสู้กับเชื้อโรคผลการตรวจสอบลำไส้เล็กด้วยกล้องจุลทรรศน์อิเล็กตรอนแบบส่องผ่านพบว่าเยื่อเมือกในลำไส้ของปลาทั้ง 2 กลุ่มมีความเสียหายเกิดขึ้นพบลักษณะเชื้อแบคทีเรียคล้ายกับ S. agalactiae ในทั้งสองกลุ่มทดลองอย่างไรก็ตาม ยังพบลักษณะเชื้อ L. plantarum เติบโตเพิ่มจำนวนปกคลุมอยู่ที่ผนังลำไส้เล็กไส้ ซึ่งช่วยเพิ่มประสิทธิภาพการเจริญเติบโตและปรับค่าตัวชี้วัดทางโลหิตวิทยาบางค่าในบทวิเคราะห์ผลของงานวิจัยเกี่ยวกับโปรไบโอติกหลายชิ้นยังพูดถึงเรื่องความเข้มข้นของเชื้อโปรไบติกมีผลกับการต่อต้านเชื้อก่อโรคอีกด้วย ถ้ามีปริมาณเชื้อโปรไบติกอยู่ในระบบทางเดินอาหารสูงจะช่วยลดปริมาณเชื้อก่อโรคในอวัยวะเป้าหมายได้ ทั้งนี้การใช้โปรไบโอติกสามารถเพิ่มความต้านทานต่อการติดเชื้อโรคในตัวสัตว์ได้โดยการเพิ่มระบบภูมิคุ้มกันที่ไม่เฉพาะเจาะจงการเลือกใช้โปรไบโอติกก็มีความสำคัญและเป็นประโยชน์ในการป้องกันโรคของฟาร์มด้วยนะคะ สัตวแพทย์หญิง ดร.มินตรา ลักขณา (หมอเม)
Probiotics and Streptococcus There were many researches, studying about the efficacy of probiotics against several diseases in aquatic animals. Streptococcosis in tilapia is among one of them. There was a study in Indonesia, testing different types and dosages of probiotics against pathogenic Streptococcus agalactiae. Three types of probiotics, Bacillus cereus, Bacillus subtillis, and Staphylococcus lentus were mixed in different combinations and dosages, and gave to the Nile tilapia with feed for 14 days. The fish were divided into 6 groups and received different combinations of probiotics as follows: 1. The fish received Bacillus cereus and Bacillus subtillis at the dosage of 1,000,000 CFU/mL. 2. The fish received Bacillus subtillis and Staphylococcus lentus at the dosage of 100,000 CFU/mL. 3. The fish received Bacillus cereus and Staphylococcus lentus at the dosage of 100,000 CFU/mL. 4. The fish received Bacillus cereus, Bacillus subtillis, and Staphylococcus lentus at the dosage of 1,000,000 CFU/mL. 5. The fish received regular feed without probiotics (positive control) 6. The fish received regular feed without probiotics and were not challenge with Streptococcus (negative control) After the fish were fed with probiotics combination for 14 days, the fish from group 1-5 were injected with virulence strain of Streptococcus agalactiae. The groups that received probiotics have higher survival rate (group 1-4, 85-89%) than the control group (group 5, 66%). In addition, immune activity of the probiotics feeding group were also higher than the control group. Another study from Brazil in 2017 showed the efficacy of probiotics Lactobacillus plantarum (lactic acid bacteria) when given with feed to Nile tilapia. The study compared growth rate and immune response of fish that received probiotics. The study also examined small intestine of the fish when challenged with Streptococcus agalactiae, using electron microscope. After 58 days of probiotics feeding, the fish those have been eaten probiotics showing more amount of good bacteria (Lactobacillus plantarum) and less amount of bad bacteria (Vibrionaceae) in their intestine, compared to the control group. The fish in probiotics-treated group also have better growth rate and better feed conversion ratio. When the fish were challenged with Streptococcus agalactiae, the fish in probiotics-treated group were found to have higher level of immune response which indicated by the higher amount of thrombocytes and neutrophils cells. Also, inside the intestine of probiotics-treated fish, they observed that the damaged intestinal wall was covered by L. plantarum. This implied that the probiotics L. plantarum helped reduce severity due to Streptococcus infection. Other researches also describe about the relationship between concentration of probiotics with pathogens invasion. If the level of probiotics in gastrointestinal tract is high, the numbers of pathogen in target organs are reduced. In addition, probiotics can stimulate the non-specific immunity which is the first line of defense against diseases.
