การเก็บตัวอย่างสัตว์น้ำเพื่อตรวจโรค
ที่ผ่านมาหลายท่านที่ติดตามเพจของทางฟาร์มนอกจากจะได้อ่านบทความเรื่องโรคและการจัดการสุขภาพสัตว์น้ำไปแล้ว น่าจะเคยมีโอกาสรับชมคลิปที่เกี่ยวกับเรื่องโรคและสุขภาพปลากันไปบ้าง เมื่อสังเกตพบลักษณะอาการที่ผิดปกติของสัตว์น้ำและสงสัยว่าป่วยควรต้องเก็บตัวอย่างสัตว์น้ำเพื่อตรวจหาสาเหตุของโรค และดำเนินการรักษา ควบคุมโรคต่อไป การเก็บตัวอย่างสัตว์น้ำทุกครั้ง จะต้องมีการจดบันทึกที่มาที่ไป ร่วมกับการซักประวัติ วิธีการเลี้ยง กรณีมีสัตว์น้ำอาการผิดปกติไปจากสภาวะปกติก็ควรต้องสังเกตลักษณะอาการและบันทึกเอาไว้ในประวัติตัวอย่างด้วย นอกจากนี้ต้องตรวจคุณภาพน้ำที่ใช้เลี้ยงปลาเพื่อนำข้อมูลมาประกอบการตัดสินใจในการดำเนินการต่อไป กรณีที่เป็นการตรวจปลาหรือกุ้งป่วยที่ฟาร์ม ส่วนใหญ่แล้วจะใช้วิธีการสังเกตพฤติกรรม อาการ รอยโรค บางฟาร์มอาจมีขั้นตอนการตรวจสุขภาพเบื้องต้นที่ฟาร์ม เช่น ตรวจพยาธิภายนอก ตรวจการกินอาหาร แต่อย่างไรก็ตามส่วนใหญ่ยังไม่สามารถดำเนินการตรวจวินิจฉัยโรคได้ครบถ้วนที่ฟาร์ม ก็จำเป็นต้องส่งตัวอย่างสัตว์น้ำป่วยเพื่อตรวจวินิจฉัยอย่างละเอียดที่ห้องปฏิบัติการหรือห้องแลปต่อไป การเก็บตัวอย่างที่ถูกวิธีมีความสำคัญมากๆ ต่อประสิทธิภาพของการวิเคราะห์และความน่าเชื่อถือของผลการตรวจที่ได้ สิ่งสำคัญนอกจากจะต้องรักษาสภาพตัวอย่างไม่ให้เน่าก่อนจะถึงห้องแลปแล้วจะต้องมีวิธีการเก็บและรักษาตัวอย่างที่ถูกต้องที่ช่วยลดการปนเปื้อนของเชื้อโรคอื่นที่อยู่ในสิ่งแวดล้อม ซึ่งถ้าเกิดมีการปนเปื้อนขึ้นมาก็อาจทำให้ผลการตรวจผิดพลาดไปได้ ตัวอย่างที่ส่งตรวจอาจเป็นไข่ปลา กุ้ง/ปลาทุกอายุ ทุกขนาดก็ได้ ควรเลือกเก็บตัวอย่างสัตว์น้ำที่สังเกตเห็นว่ามีอาการป่วย มีรอยโรค ร่วมกับการเลือกเก็บสัตว์น้ำในกลุ่มเดียวกันที่ไม่พบอาการป่วย จะเป็นประโยชน์ในการเปรียบเทียบผลการตรวจ และวินิจฉัยสภาวะความรุนแรงของโรคในฟาร์ม สำหรับปริมาณตัวอย่างที่เก็บควรพิจารณาร่วมกับความชุกของโรคถ้าพบความชุกมาก อาจเก็บตัวอย่างไม่มาก เนื่องจากมีโอกาสพบสาเหตุของโรคได้มากกว่า แต่ถ้ามีความชุกของโรคน้อยต้องพิจารณาเก็บตัวอย่างปริมาณมากขึ้นเพื่อให้เพิ่มโอกาสในการตรวจพบสาเหตุของโรค
ยกตัวอย่างขั้นตอนในการเก็บตัวอย่างปลาเพื่อส่งตรวจโรคในห้องแลป ดังนี้ นะคะ - ถ้าเป็นปลามีชีวิตก็สุ่มมาพร้อมกับน้ำในบ่อที่เลี้ยงด้วยเลย ใส่ในภาชนะที่เหมาะสม เช่น ถุงพลาสติก ถังน้ำ โดยแยกตัวอย่างแต่ละบ่อเอาไว้ไม่ให้ปะปนกัน และเขียนระบุรายละเอียดให้ชัดเจนว่าเป็นตัวอย่างจากบ่อไหน จากนั้นจัดการขนส่งมาที่ห้องแลป - กรณีที่ไม่สามารถส่งปลาที่มีชีวิตมาตรวจได้ นำตัวอย่างปลาที่เพิ่งตายใส่ในถุงพลาสติก ปิดปากถุงให้สนิทแช่น้ำแข็ง แล้วรีบส่งที่ห้องแลปทันทีหรือภายในวันที่เก็บตัวอย่าง แต่ถ้าส่งไม่ได้ควรแช่แข็งปลาทั้งตัวในน้ำแข็งแห้งหรือตู้แช่ที่อุณหภูมิลบ 20 ºC หรือต่ำกว่า แล้วในวันต่อมาให้รีบส่งที่ห้องแลป - แต่ถ้าหากไม่สามารถนำตัวอย่างปลามีชีวิตหรือตัวอย่างที่แช่แข็งส่งห้องแลปได้ทันที ให้ใช้เอทานอล 90% - 95% ในการรักษาสภาพตัวอย่าง กรณีที่ปลาตัวเล็กน้ำหนักน้อยกว่า 5 กรัม ก็สามารถเอาปลาทั้งตัวแช่ลงไปในเอทานอล 90% - 95% ได้เลย แต่ถ้าหากว่าปลามีขนาดใหญ่มาก แนะนำให้เลือกเก็บตัวอย่างอวัยวะภายในของปลาที่เป็นอวัยวะเป้าหมายที่เชื้อชอบเจริญเติบโต ก็คือ ไต สมอง และม้าม โดยผ่าซากปลาและแยกเก็บอวัยวะเป้าหมายแช่ลงไปในเอทานอล 90% - 95% ปริมาตร 10 เท่าของขนาดตัวอย่าง แล้วเก็บตัวอย่างไว้ในตู้เย็น แล้วในวันต่อมาให้รีบส่งที่ห้องแลปเพื่อตรวจ การตรวจโรคจะช่วยในการวินิจฉัยหาสาเหตุของการเกิดโรคในฟาร์มเพื่อการวางแผนจัดการด้านสุขภาพ และควบคุมป้องกันโรคระบาดและช่วยให้ทราบสภาวะของโรคภายในฟาร์มนะคะ สัตวแพทย์หญิง ดร.มินตรา ลักขณา (หมอเม)
Sample collection for disease diagnosis If you have been following our Facebook page, you must have read about diseases and health management in aquatic animals. Or, you might have a chance to watch some videos about diseases or fish health management. Have you ever wonder? How do we collect samples when the fish show abnormal behavior or signs of illness? Good sample collection leads to accurate disease diagnosis which will lead to the effective treatment and control. Every time you collect samples, always include history taking and record keeping on the important details such as culture system and abnormal signs or clinical signs of the animals. Important! Water from the same pond should also be collected. The water quality data is useful for the decision making to fix the ongoing problem. If the health fish or shrimp were assessed on site, primary evaluation can only be performed such as observation of behavior or external lesion, feeding rate, and screening for ectoparasite; however, the real problem cannot be identified and confirmed. In other words, disease confirmation requires the sample to be sent to the laboratory for better and more accurate diagnostic methods. Appropriate sample collection technique is very important to the efficacy and reliability of the diagnostic procedure. Besides maintaining the freshness of the samples, you must prevent contamination by other pathogens in the environment which will lead to misdiagnosis. Samples can be anything from fish eggs, shrimp/fish at all age and size. The best is to correct moribund fish that shows clinical lesion and also collect fish in the same pond that clinical sign is absent (to help in result interpretation). The severity of the disease outbreak on the farm should be recorded such as mortality, and morbidity rate. The number of samples to be collected depends on disease prevalence. Higher prevalence requires less number of samples and vice versa. These are the following sample collection for laboratory testing: - Live fish should be collected with the pond water. Keep sample in an appropriate container such as plastic bag or bucket. Separate sample from each pond and label the samples clearly. - If live fish cannot be obtained, collect the fish that just died and keep it in a plastic bag. Tightly close the bag and store in ice. Send the sample to the laboratory immediately or within the same day of sample collection. If the sample cannot be sent, keep the whole fish in plastic bag and store in dry ice or in a freezer at -20 oC or lower. The sample should be submitted to the laboratory on the next day. - If the 2 methods described above cannot be performed, preserve the sample in 90-95% ethanol. For small fish (less than 5 g), submerge the whole fish in ethanol. For bigger size fish, dissect target organs of pathogens such as kidney, brain, and spleen, and submerge those organs in ethanol. The volume of ethanol should be approximately 10 times more than the samples. Disease diagnosis will help you identify the cause of a disease that occurs in your farm.
