การสร้างความสมดุลของภูมิคุ้มกัน ตามที่เขียนไปแล้วเมื่อครั้งก่อนว่า ระบบภูมิคุ้มกันของสัตว์น้ำจะมีการพัฒนาและมีความซับซ้อนน้อยกว่าสัตว์มีกระดูกสันหลังชนิดอื่น นอกจากนี้ระบบภูมิคุ้มกันในสัตว์น้ำวัยอ่อนก็ยังพัฒนาได้น้อยกว่าสัตว์น้ำที่โตเต็มวัยและมักจะพบการตอบสนองทางภูมิคุ้มกันแบบไม่จำเพาะในสัตว์น้ำวัยอ่อนมากกว่าการเกิดภูมิคุ้มกันแบบจำเพาะ ปัจจุบันมีงานวิจัยที่แสดงให้เห็นว่าการให้สัตว์น้ำวัยอ่อน ลูกปลากินโปรไบโอติกแบคทีเรียหรือใส่โปรไบโอติกในบ่อเลี้ยงสัตว์น้ำจะช่วยกระตุ้นให้เกิดภูมิคุ้มกันแบบไม่จำเพาะได้และยังพบว่าปริมาณโปรไบโอติกแบคทีเรียยึดเกาะพื้นที่ในลำไส้แปรผันตามปริมาณของโปรไบโอติกแบคทีเรียที่ผสมในอาหารให้สัตว์น้ำกินซึ่งปกติแล้วจุลินทรีย์ประจำถิ่น (microflora) ที่อยู่ในลำไส้มีประสิทธิภาพในการช่วยป้องกันการติดเชื้อ ผลิตสารที่มีประโยชน์ช่วยในการย่อยและดูดซึมสารอาหารในระบบทางเดินอาหารของปลาอยู่แล้วแต่การนำโปรไบโอติกแบคทีเรียมาใช้ในอาหารจะช่วยเสริมประสิทธิภาพการแย่งพื้นที่ยึดเกาะกับเชื้อก่อโรค ซึ่งการนำโปรไบโอติกมาใช้ในอุตสาหกรรมการผลิตสัตว์น้ำก็ได้รับการยอมรับให้ใช้เพิ่มมากขึ้น โดยเล็งเห็นว่าจะช่วยควบคุมเชื้อก่อโรคในสัตว์น้ำ มีหลายการศึกษาวิจัยที่พยายามหาคำตอบเกี่ยวกับกลไกการทำงานที่แตกต่างกันของโปรไบโอติกแบคทีเรียที่มีผลต่อระบบภูมิคุ้มกันซึ่งรวมถึงประสิทธิภาพการกระตุ้นให้เซลล์ในระบบภูมิคุ้มกันสังเคราะห์และหลั่งสารโปรตีนที่มีผลต่อการทำงานของระบบภูมิคุ้มกัน (cytokine) โดย cytokine มีผลต่อการกระตุ้นภูมิคุ้มกันที่ทำงานหลายๆ อย่างในร่างกายเช่น กระตุ้นหรือยับยั้งการอักเสบ ต้านไวรัส กระตุ้นการแบ่งเซลล์ในระบบภูมิคุ้มกันหลังจากการติดเชื้อ ซ่อมแซมเนื้อเยื่อ และยังกระตุ้นภูมิคุ้มกันในรูปแบบอื่นๆ นอกเหนือจากที่กล่าวมาตามความเหมาะสม นอกจากโปรไบโอติกแบคทีเรียจะยับยั้งการเกาะกลุ่มกันของเชื้อแบคทีเรียก่อโรคในระบบทางเดินอาหารหรือแย่งพื้นที่ยึดเกาะของเชื้อก่อโรคในผนังลำไส้ของสัตว์แล้วโปรไบโอติกแบคทีเรียแต่ละชนิดยังมีประสิทธิภาพต้านทานเชื้อก่อโรคต่างกันโดยขึ้นอยู่กับระบบภูมิคุ้มกันของสัตว์น้ำแต่ละชนิดและพยาธิกำเนิดของเชื้อก่อโรคแต่ละชนิดด้วย มีการศึกษาผลของโปรไบโอติกแบคทีเรียต่อการตอบสนองทางภูมิคุ้มกันปริมาณเชื้อแบคทีเรียก่อโรคในตัวสัตว์รวมทั้งสภาพแวดล้อมเอาไว้อยู่หลายการศึกษา เช่น การให้ปลาเทราท์กินโปรไบโอติกแบคทีเรีย Clostridium butyricum ทำให้เพิ่มความทนทานต่อการติดเชื้อก่อโรควิบริโอ (vibriosis) โดยพบว่าเม็ดเลือดขาวมีประสิทธิภาพการเก็บกินเชื้อก่อโรคเพิ่มมากขึ้นการผสมโปรไบโอติกแบคทีเรียลงในอาหารหรืออาหารที่มีชีวิต (live food) หรือใส่ลงในน้ำที่เลี้ยงช่วยกระตุ้นภูมิคุ้มกันแบบไม่จำเพาะและมีผลช่วยลดปริมาณเชื้อแบคทีเรียในปลากระดูกแข็งหลายชนิด เช่น ปลาแซลมอน ปลา turbot (Scophthalmus maximus) นอกจากนี้ยังมีรายงานว่าโปรไบโอติกแบคทีเรียกลุ่ม lactic acid สามารถหลั่งสาร hydrogen peroxide และสาร bacteriocin-like substances ออกมาซึ่งมีคุณสมบัติในการยับยั้งเชื้อแบคทีเรียแกรมลบ Vibrio metschnikovii และ V. harveyi และแกรมบวกคือเชื้อ Staphylococcus aureus ที่ก่อโรคในปลาเก๋าจุดน้ำตาล หรือ ปลากะรังปากแม่น้ำ (Orange-spotted grouper) นอกจากนี้ในปลากลุ่มที่ทดลองให้กินโปรไบโอติกแบคทีเรีย Bacillis spp. ยังพบระดับของภูมิคุ้มกันไม่จำเพาะเพิ่มสูงขึ้น เช่นประสิทธิภาพในการเก็บกินและทำลายเชื้อก่อโรคเพิ่มขึ้น มีการทดลองนำเชื้อ Bacillus spp. ผสมอาหารให้กุ้งขาวกินพบว่ามีผลต่ออัตราการเจริญเติบโตและการรอดชีวิตเพิ่มขึ้น และพบว่าป้องกันเชื้อก่อโรค V. harveyi และเชื้อไวรัสจุดขาว (white spot syndrome virus) ซึ่งโปรไบโอติกแบคทีเรียจะกระตุ้นระบบภูมิคุ้มกันโดยเพิ่มประสิทธิภาพการเก็บกินเชื้อก่อโรคและการยับยั้งเชื้อแบคทีเรียเมื่อตรวจวัดค่าภูมิคุ้มกันโดยพารามิเตอร์ต่างๆ เช่น phenoloxidase, prophenoloxidase (proPO), respiratory bursts และ superoxide dismutase activity รวมทั้งประสิทธิภาพในการทำลายเชื้อพบว่ามีประสิทธิภาพดีขึ้นในปลากลุ่มที่ได้รับโปรไบโอติกแบคทีเรียผสมอาหาร - ภูมิคุ้มกันแบบไม่จำเพาะ เป็นกลไกในการกำจัดสิ่งแปลกปลอมที่เข้ามาในร่างกายแบบไม่จำเพาะเจาะจง (ไม่ระบุชนิด) มีประสิทธิภาพในการป้องกันทำลายสิ่งแปลกปลอมหรือเชื้อจุลินทรีย์ได้เพียงระดับหนึ่งเท่านั้น - ภูมิคุ้มกันแบบจำเพาะหรือการตอบสนองของระบบภูมิคุ้มกัน (immune response) เป็นกลไกกำจัดสิ่งแปลกปลอมในร่างกายที่มีความจำเพาะต่อสิ่งแปลกปลอมนั้น เช่น เชื้อจุลินทรีย์ เซลล์ หรือสิ่งที่เกิดขึ้นจากความผิดปกติในร่างกาย
From last time. We mentioned that immune system in fish is less developed and more primitive than other vertebrates. In addition, immune system of younger fish is also less developed than mature fish. Younger fish relies mostly on “non-specific” immune response, rather than “specific” one. Recently, there are several researches showing immunostimulating effects of probiotics when given to juvenile fish or fingerling. Scientists also found that number of microflora in gut associates with feeding juvenile fish with probiotics. What are “microflora”? Microflora are good bacteria living in animal’s gut. They colonize in animal’s intestine and help prevent infestation of bad microorganisms. Microflora also produce substances that support digestion and absorption of nutrients. By giving probiotics with feed, it will help the microflora in occupying gut’s surface area, so bad bacteria cannot colonize. There are increasing uses of probiotics in aquaculture. The main idea is to control diseases. Many researchers are trying to figure out how probiotics enhance immune system. Also, how do probiotics stimulate the production of important immune-related substances, as known as Cytokines. Cytokines play important role in immune system. For example, they regulate inflammatory response, involve in the fight against viruses, activate cell replication, involve in tissue repair, etc.
Not only that probiotics can inhibit the attachment of bad bacteria in gut, probiotics themselves have antimicrobial property. There is a study showing that white blood cells of trout have better performance against Vibrio spp. when fed with probiotics Clostridium butyricum. By mixing probiotics with live feed given to in salmon and turbot showed that it helps stimulate non-specific immune response and reduce number of bad bacteria. Another type of bacteria, Lactic acid bacteria (or LAB) are commonly used as probiotics. These groups of bacteria can produce hydrogen peroxide and bacteriocin-like substances which can inhibit the growth of several bad bacteria, such as Vibrio metschnikovii, V. harveyi and Gram-positive bacteria, Staphylococcus aureus, an important pathogen causing disease in orange-spotted grouper. Another famous probiotics bacterium, Bacillis spp., also can stimulate non-specific immune response, such as engulfing and killing of pathogens (phagocytosis). Bacillis bacteria were given to Pacific white shrimp (Litopenaeus vannamei) and the result showed that the shrimp obtained better growth rate and higher survival rate when challenged with V. harveyi and white spot syndrome virus. It was found that the immune response of shrimp was stimulated after receiving probiotics and several immune parameters, such as phenoloxidase, prophenoloxidase (proPO), respiratory bursts and superoxide dismutase activity were found to be higher compare to the group that did not receive probiotics. - Non-specific immunity: first responder, immune response that come first and try to eliminate any pathogen invading the body. This type of immunity is less effective than the specific immunity. - Specific immunity: another type of immune response. This type of response is specific to each particular pathogen (different species of bacteria, virus, etc.). Due to this specificity, this type of immunity can eliminate pathogen much more effective than the non-specific immunity. |
